Thinking back t the material I presented early on, I used "tier" from our internal (Vnomic specific) vernacular, which is defined as a set of homogenous nodes capable of hosting specific kinds of software, which is purely a topological concept useful for grouping, and concisely defines kind (the type of node you can have) and cardinality. We have also defined clusters to be a sub type of tiers with more specific behavioral semantics in terms of availability, scalability and placement. The most common kind of thing we host on a tier is a stack which is often considered layered. "Stack" seems to provide or imply layering, but I think we would use "stack" only when we want to refer to specific sets of functionality. Layering seems relevant when we talk about where certain functions are provided, but in terms of abstraction. So I guess I'd say that tier is a topological concept whereas layer is a functional abstraction concept and they generally are orthogonal. 

Such fundamental concepts are certainly worthy of specific attention and concise definition. Thinking back, this is precisely why we decided to define tier as a first class member of our domain specific vocabulary... At the very least we could add a glossary section with these terms somewhere.
